Uno studio, a cura della Microbiologia del DIMEC UniBo e  del Laboratorio Unico della Romagna, pubblicato sulla rivista “Nature Vaccines”, ipotizza un nuovo vaccino contro il Sars- Cov-2 a basso costo e ad alta efficacia

Il paper, a cura dell’équipe di Microbiologia del Laboratorio Unico dell’Ausl Romagna, diretta dal prof. Vittorio Sambri, intitolato “Development and in vivo evaluation of a SARS-CoV-2 inactivated vaccine using high hydrostatic pressure” e pubblicato su NPJ Vaccines il 25 aprile 2025

(https://www.nature.com/articles/s41541-025-01136-7), presenta “una nuova strategia per la produzione di vaccini inattivati contro SARS-CoV-2 utilizzando la tecnologia della pressione idrostatica elevata (HHP)”.

“L’obiettivo principale – spiega il prof. Sambri – è validare l’uso della HHP come metodo economico, rapido e scalabile per produrre vaccini a virus intero inattivati, mantenendo un’elevata immunogenicità e stabilità strutturale del vibrione, caratteristiche fondamentali per ottenere un preparato vaccinale office e con alta possibilità di  essere usato in contesti con risorse economiche e strutturali limitate”

Qual è stato il metodo utilizzato dal team di ricerca e che risultati sono stati ottenuti?

“Il virus SARS-CoV-2 – spiega il prof. Sambri –  è stato sottoposto a diversi livelli di pressione (fino a 600 MPa). È stato osservato che pressioni più elevate (500–600 MPa) causano una destabilizzazione della morfologia virale. In modelli animali (questa parte dello studio è stata eseguita presso l’istituto Zooprofilattico Sperimentale della Lombardia Emilia a Brescia) il virus, trattato a 500 Mpa, ha indotto le risposte immunitarie umorali e cellulari più robuste, superando di gran lunga i risultati eseguiti con virus inattivato  termicamente.  Il metodo, in pratica, lascia intatta la capacità del virus di indurre una risposta verosimilmente protettiva (almeno nel modello animale)  mentre lo stesso virus perde totalmente la capacità di infettare. Inoltre, il  vaccino inattivato con HHP ha mantenuto la stabilità per 30 giorni a 4 °C, riducendo la dipendenza dalla catena del freddo.”

“I principali risultati – conclude il prof Sambri, che è anche docente ordinario di Microbiologia al Dipartimento di Scienze mediche e Chirurgiche all’Università di Bologna –  sono stati l’efficacia immunitaria (il trattamento a 500 MPa ha generato risposte immunitarie superiori rispetto all’inattivazione termica),  la stabilità del vaccino (la preparazione vaccinale ha mantenuto la sua efficacia per un mese a 4 °C, facilitando la distribuzione in aree con infrastrutture limitate) e , infine, la scalabilità e i costi: la  tecnologia HHP offre un processo di produzione rapido, economico e facilmente scalabile, ideale per affrontare pandemie emergenti.

L’adozione della tecnologia HHP per la produzione di vaccini potrebbe rappresentare una svolta significativa, soprattutto per i paesi a basso e medio reddito, grazie alla riduzione dei costi e alla facilità di distribuzione senza necessità di catene del freddo complesse.”
